Transcendis biotechnology improves naked mRNA transfection of mouse skin, skeletal muscle or tumor.

Naked uncapped mRNA possesses a peptide tag-binding RNA aptamer at its 5’end.

A transfection peptide possesses this tag at its C-terminal end.

Naked mRNA and the transfection peptide are mixed and allowed to bind to each other at room temperature for a few minutes.

A single copy of the transfection peptide non-covalently and strongly binds to mRNA.

The injection of such transfection peptide/mRNA system to mouse skin leads to a 9.3-fold enhancement of protein expression compared to mRNA alone.

Thus, Transcendis greatly improves the uptake of naked mRNA by skin cells.
